DESCRIPTION

MART-1 (26–35) is the native human Melan-A/MART-1 decamer EAAGIGILTV. Together with the overlapping MART-1 (27–35) nonamer, it represents a major HLA-A*02:01-associated antigenic region recognized by melanoma-reactive cytotoxic T cells.

The additional N-terminal glutamic acid distinguishes this peptide from the 27–35 nonamer and makes the two sequences particularly useful as a matched system for studying peptide length, MHC presentation, and T-cell receptor recognition.

Native Decamer versus Modified MART-1 Peptides

MART-1 (26–35) has been extensively compared with the anchor-modified sequence ELAGIGILTV, in which the second residue is changed from alanine to leucine.

That substitution improves HLA-A2 binding, but structural and functional studies have shown that changes intended to strengthen MHC anchoring can also alter the way a TCR encounters the peptide–MHC complex. For this reason, the native EAAGIGILTV sequence remains an important control in studies evaluating modified MART-1 ligands.
