MART-1 (26–35) is the native human Melan-A/MART-1 decamer EAAGIGILTV. Together with the overlapping MART-1 (27–35) nonamer, it represents a major HLA-A*02:01-associated antigenic region recognized by melanoma-reactive cytotoxic T cells.
The additional N-terminal glutamic acid distinguishes this peptide from the 27–35 nonamer and makes the two sequences particularly useful as a matched system for studying peptide length, MHC presentation, and T-cell receptor recognition.

Native Decamer versus Modified MART-1 Peptides
MART-1 (26–35) has been extensively compared with the anchor-modified sequence ELAGIGILTV, in which the second residue is changed from alanine to leucine.
That substitution improves HLA-A2 binding, but structural and functional studies have shown that changes intended to strengthen MHC anchoring can also alter the way a TCR encounters the peptide–MHC complex. For this reason, the native EAAGIGILTV sequence remains an important control in studies evaluating modified MART-1 ligands.
Comparative Experimental Design
Researchers can pair AS2704 with MART-1 (27–35), Human to investigate nonamer versus decamer presentation while keeping the central MART-1 sequence largely conserved.
Native, anchor-modified, or systematically substituted variants can also be prepared through Custom Peptide Synthesis.
Analytical Considerations
For peptide–MHC and T-cell assays, purity requirements should be matched to the experimental system. HPLC purity and mass confirmation provide useful complementary information when comparing native and modified sequences.
Additional guidance is available in our Recommended Peptide Purity resource.
Storage and Handling
Store the lyophilized material according to the supplied batch documentation. For repeated experiments, suitable aliquoting can help maintain consistent handling between assay runs.
Research Use Only
