H-Ser(tBu)-OH is O-tert-butyl-L-serine. The serine side-chain hydroxyl is protected as a tert-butyl ether, while the α-amino and α-carboxyl groups remain unprotected.
This protection state is useful as a precursor when a route requires control over the serine hydroxyl but needs a different N-protecting group, ester, salt, or derivatization sequence than standard preassembled Fmoc-Ser(tBu)-OH.

Selective Side-Chain Protection
The tBu group masks the hydroxyl and reduces undesired O-acylation or other side reactions during transformations at the amino or carboxyl group. The α-amino group can then be converted to Fmoc, Boc, Cbz, an amide, or another route-specific derivative.
Because the carboxyl group remains free, the material can also be esterified or activated according to the target intermediate.
When to Use H-Ser(tBu)-OH Instead of Fmoc-Ser(tBu)-OH
Choose the free-amino precursor when the N-protection must be introduced in-house or when a solution-phase route does not use Fmoc chemistry. For routine automated Fmoc-SPPS, a pre-Fmoc-protected serine derivative is usually more direct.

Identity and Deprotection Planning
The tBu ether is acid-labile under sufficiently strong conditions. Deprotection should be planned with the full molecule because acid-sensitive groups elsewhere in the route may limit conditions.
