Precursor Form Versus Mature GLP-1

The potent incretin forms GLP-1 (7-36) amide and GLP-1 (7-37) begin six residues downstream of the N-terminus of GLP-1 (1-37).

This processing difference has major functional consequences. Classical pancreatic perfusion experiments found GLP-1 (1-37) to be far less potent for insulin secretion than mature GLP-1 (7-37).

We recommend using GLP-1 (1-37) primarily when precursor processing or noncanonical biology is the experimental question, not as an interchangeable replacement for mature GLP-1.

Distinct Experimental Biology

Beyond conventional incretin assays, GLP-1 (1-37) has also been investigated in intestinal epithelial-cell models, where specific effects have been reported that differ from its weak acute insulinotropic profile.

This creates a useful example of how different proglucagon-derived molecular forms can have different biological contexts despite sharing most of their sequence.
