DESCRIPTION

Exendin-4 is a 39-residue peptide agonist of the glucagon-like peptide-1 receptor (GLP-1R) originally isolated from Heloderma suspectum. Its sequence combines a receptor-activating N-terminal region with a C-terminal Exendin-specific extension that contributes to ligand affinity and conformational behavior.

Because it is structurally distinct from endogenous GLP-1 while activating the same receptor system, Exendin-4 is widely used as a reference ligand in class B1 GPCR pharmacology.

Distinct Engagement of GLP-1R

Exendin-4 and GLP-1 both activate GLP-1R, but they do not engage the receptor identically. Structural and kinetic work has shown differences in N-terminal receptor contacts, ligand dynamics and coupling to G-protein activation.

This makes Exendin-4 valuable for studies focused on ligand-specific receptor conformations, signaling kinetics and biased or pathway-dependent responses.

DPP-4 Resistance and Experimental Stability

Native GLP-1 is rapidly cleaved by DPP-4 at its N-terminus. Exendin-4 has different N-terminal sequence chemistry and is substantially more resistant to this route of degradation.

For kinetic studies, this distinction should be considered when Exendin-4 and native GLP-1 are compared over extended incubation periods.
