Fmoc-D-His(Trt)-OH is an Fmoc-protected D-histidine building block with the imidazole side chain protected by trityl. The protecting-group combination is compatible with standard Fmoc/tBu solid-phase peptide synthesis and keeps the imidazole protected through iterative deprotection and coupling steps.
Histidine requires additional stereochemical attention because activated His derivatives can undergo alpha-carbon epimerization under some coupling conditions. For D-His-containing peptides, chiral purity of the starting material and coupling conditions both affect the final stereochemical integrity.
