Fmoc-D-Asn(Trt)-OH is an Fmoc-protected D-asparagine derivative used in Fmoc solid-phase peptide synthesis. The alpha-amino group is protected with Fmoc and the side-chain amide is protected with trityl (Trt), which remains intact during base-mediated Fmoc removal and is cleaved under acidic final-deprotection conditions.
This building block is useful when D-asparagine must be introduced with controlled side-chain protection. It should be distinguished from Fmoc-D-Gln(Trt)-OH: both are Trt-protected amide residues, but glutamine contains one additional methylene group and therefore has a different formula, molecular weight and CAS number.

Application Scope and Assay Relevance
Primary use: Protected D-asparagine building block for stereochemically defined peptide synthesis rather than a direct bioassay reagent.
| Typical in vitro relevance | After peptide synthesis, D-Asn substitutions can be assessed in receptor- or protein-binding assays, enzyme inhibition, protease/serum stability and conformational SAR. The main experimental value is controlled inversion of local stereochemistry while retaining an amide side chain. |
| In vivo relevance | In vivo relevance belongs to the final peptide sequence. D-Asn incorporation can be part of a metabolic-stability strategy, but no general in vivo benefit should be inferred without sequence-specific validation. |
Asn(Trt) vs Gln(Trt)
The two protected amide residues look similar in abbreviated naming, but they differ by one side-chain methylene group. CAS number and molecular weight are useful checks against picking the wrong building block.
Purchasing and QC Considerations
Review CAS identity, chiral purity, HPLC purity, water content and lot COA. Supplier nomenclature may use slightly different Greek-position conventions for the Trt-protected amide; the product shorthand and CAS are the more robust identifiers.
Frequently Asked Questions
What does Trt protect in this building block?
Trt protects the D-asparagine side-chain amide.
Is Trt removed during Fmoc deprotection?
No. It normally remains during base-mediated Fmoc cycles and is removed during acidic final cleavage.
How is it different from Fmoc-D-Gln(Trt)-OH?
D-Gln has one additional side-chain methylene group and a different CAS and molecular weight.
