Fmoc-D-Gln(Trt)-OH is an Fmoc-protected D-glutamine building block for solid-phase peptide synthesis. The alpha-amino group carries Fmoc protection and the glutamine side-chain amide is protected with Trt, allowing standard base-mediated Fmoc cycles while retaining side-chain protection until final acidic cleavage.
It is used in partially D-configured peptides, all-D sequences and stereochemical SAR projects requiring D-Gln. Because Asn(Trt) and Gln(Trt) names are visually similar, users should verify the CAS number and molecular weight; Fmoc-D-Gln(Trt)-OH contains one additional methylene group relative to the corresponding D-Asn derivative.

Application Scope and Assay Relevance
Primary use: Protected D-glutamine building block for D-peptide synthesis and stereochemical peptide optimization.
| Typical in vitro relevance | D-Gln substitutions are most relevant to binding, enzyme-inhibition, protease/serum-stability and conformational SAR assays after the final peptide is synthesized and deprotected. They allow local chirality to be changed while preserving a neutral amide side chain. |
| In vivo relevance | The final peptide may be evaluated in vivo if the D-Gln substitution preserves the required target activity. Metabolic stability can improve with D-residue incorporation, but the outcome is sequence-specific. |
Gln(Trt) vs Asn(Trt)
D-Gln and D-Asn are homologous amide residues but are not interchangeable. The protected D-Gln reagent has a higher molecular weight because of the additional side-chain methylene group.
Purchasing and QC Considerations
Supplier naming conventions for the side-chain amide nitrogen can vary. Use the product shorthand, CAS 200623-62-7, formula and MW as the primary identity checks, together with chiral purity and COA.
Frequently Asked Questions
What does Trt protect?
Trt protects the glutamine side-chain amide.
When is Trt removed?
It is normally removed during acidic final cleavage.
How does D-Gln(Trt) differ from D-Asn(Trt)?
D-Gln contains one additional methylene group and has a different CAS number and molecular weight.
