Fmoc-D-Arg(Pbf)-OH is an Fmoc-protected D-arginine derivative for solid-phase peptide synthesis. The alpha-amino group is protected with Fmoc, while the guanidino side chain is protected by Pbf, an acid-labile protecting group widely used in Fmoc/tBu peptide chemistry.
Pbf protection suppresses unwanted guanidino-side-chain reactions during iterative coupling and deprotection and is removed during final acidic cleavage. This building block is useful for introducing D-arginine into cationic peptides, stereochemical analogs and D-amino-acid libraries.

Application Scope and Assay Relevance
Primary use: Building block for D-arginine incorporation into cationic, cell-penetrating, antimicrobial and other D-amino-acid-containing peptides.
| Typical in vitro relevance | Common downstream assays include live-cell uptake by fluorescence microscopy or flow cytometry, cargo-delivery studies, antimicrobial MIC assays, membrane-permeabilization assays, hemolysis/cytotoxicity testing, and trypsin/plasma-protease stability. D-arginine-rich peptides are especially relevant to protease-resistant cell-penetrating peptide design. |
| In vivo relevance | Final D-Arg-containing peptides may be evaluated for pharmacokinetics, tissue delivery, efficacy and toxicity. In vitro improvement should not be assumed to translate directly in vivo; D-amino-acid-substituted antimicrobial peptides have shown substantial differences between in vitro and animal results. |
Why Pbf is used on D-arginine
Arginine’s guanidino group is strongly basic and highly functionalized. Pbf masks it during synthesis while remaining compatible with standard Fmoc deprotection and TFA-based global cleavage.
Purchasing and QC Considerations
Use the protected-building-block molecular weight for solution and coupling calculations. Confirm CAS identity, HPLC purity, enantiomeric purity and water content for long or multi-D-residue sequences.
Frequently Asked Questions
What does Pbf protect?
Pbf protects the D-arginine guanidino side chain.
When is Pbf removed?
It is typically removed during final acidic cleavage and global deprotection.
