DESCRIPTION

Neurokinin B (NKB) is a 10-residue endogenous tachykinin and the principal endogenous agonist of the neurokinin-3 receptor (NK3R). The mature sequence is Asp-Met-His-Asp-Phe-Phe-Val-Gly-Leu-Met-NH₂.

Beyond classical tachykinin receptor pharmacology, NKB has become an important molecular tool for investigating hypothalamic KNDy neuronal networks and regulation of pulsatile reproductive neuroendocrine signaling.

Principal Endogenous NK3 Receptor Ligand

NKB can interact with all three major tachykinin receptors, but its highest endogenous receptor preference is generally associated with NK3R. Human pharmacological data show substantially stronger NK3 activity than NK1 activity.

For subtype-specific experiments, receptor expression and species should still be documented because tachykinins display overlapping pharmacology.

NKB in the KNDy Network

Neurokinin B is coexpressed with kisspeptin and dynorphin in hypothalamic KNDy neurons. Current evidence supports NKB-NK3R signaling as an important stimulatory component involved in synchronization of this neuronal network and regulation of pulsatile GnRH output.

This makes the peptide relevant not only to conventional tachykinin pharmacology but also to reproductive neuroendocrine and hypothalamic circuit research.

NKB contains Met2 and Met10. Oxidation of either residue can generate molecular species with altered mass and potentially altered receptor behavior.
