DESCRIPTION

Melanocyte Associated Antigen gp100 is a transmembrane glycoprotein primarily expressed in melanocytes and melanoma cells, serving as a critical tumor-associated antigen in the immune response against melanoma. Also known as PMEL or silver protein, gp100 contains multiple antigenic epitopes that are recognized by cytotoxic T lymphocytes (CTLs) when presented by major histocompatibility complex (MHC) class I molecules. These epitopes, such as gp100 (209-217) and gp100 (178-187), are pivotal in immunotherapeutic strategies, including cancer vaccines and adoptive T-cell therapies, designed to stimulate or enhance CTL-mediated killing of melanoma cells. While gp100 is essential for melanin biosynthesis in normal melanocytes, its overexpression in melanoma makes it a target for immune-based interventions. Research focuses on optimizing gp100-derived peptides for improved immunogenicity, understanding their interaction with MHC molecules, and integrating them into combination therapies to boost anti-tumor responses, highlighting its significance in both melanoma diagnosis and treatment.
