MART-1 (27–35) is the native 9-amino-acid sequence AAGIGILTV derived from human MART-1/Melan-A (MLANA). It is one of the best-characterized melanoma-associated HLA-A2-restricted T-cell epitopes and has been recognized by melanoma-reactive tumor-infiltrating lymphocytes in multiple experimental systems.
A useful feature of this native nonamer is that its HLA-A*02:01 binding is not optimized: alanine occupies the second peptide position rather than a stronger canonical HLA-A2 anchor residue. This has made MART-1 (27–35) particularly informative for studying the relationship among MHC binding, peptide conformation, and T-cell receptor recognition.

A Native MART-1 Reference Epitope
MART-1 (27–35) is valuable as a native-sequence reference, especially when comparing altered peptide ligands designed to increase HLA-A2 binding.
For example, substitution of the second alanine with leucine produces ALGIGILTV, which binds HLA-A2 more strongly but can alter T-cell recognition. This distinction illustrates why improved MHC affinity does not necessarily translate into equivalent TCR recognition.
