IL-6R Shedding-Site Sequence

The TSLPVQDSSSVP sequence encompasses the membrane-proximal region of human IL-6R associated with receptor ectodomain shedding.

Mass-spectrometric studies of soluble human IL-6R have localized physiologically relevant proteolysis between Pro355 and Val356. This cleavage region is processed by ADAM17 and is also relevant to ADAM10-dependent receptor shedding.

The serine-rich sequence downstream of the cleavage region contributes to the local physicochemical environment of the receptor stalk and has been investigated in studies of protease recognition.

Why IL-6R Shedding Matters

Proteolytic release of membrane IL-6R generates soluble IL-6R, which can participate in IL-6 trans-signaling through gp130-expressing cells.
