DESCRIPTION

H-Leu-AMC·HCl couples L-leucine to 7-amino-4-methylcoumarin (AMC) and is supplied as the hydrochloride salt. Cleavage of an appropriate Leu–AMC amide can release the fluorescent AMC reporter, making this chemistry useful in enzyme-assay design.

For a different aromatic reporter format, Alan Scientific also lists H-Leu-βNA·HCl, which uses β-naphthylamide rather than AMC.

Application Scope in Fluorogenic Protease Assays

Leu-AMC chemistry is used to develop substrates for aminopeptidases and proteases that recognize a Leu-containing cleavage site. It can support kinetic assay optimization, enzyme-activity screening, inhibitor testing and substrate-specificity studies when the enzyme system is appropriate.

Reporter Chemistry and Signal Generation

AMC is weakly fluorescent while amide-bound relative to free reporter in many assay formats; enzymatic release increases the measurable signal. Excitation/emission settings, buffer pH, substrate concentration and enzyme specificity should be validated for the actual assay.

Confirm CAS 62480-44-8, C16H21ClN2O3, MW 324.81 g/mol and HCl salt state. For assay work, purity, free AMC background and lot-to-lot fluorescence baseline can be more important than nominal mass alone.
