Home Product Unusual Amino Acids & Analogs H-Glu(OBzl)-OtBu·HCl

DESCRIPTION

H-Glu(OBzl)-OtBu·HCl is a doubly ester-protected L-glutamate derivative in which the α-carboxyl group is protected as a tert-butyl ester and the side-chain γ-carboxyl group is protected as a benzyl ester. The amino group is supplied as the hydrochloride salt.

This asymmetric protection pattern is useful when the two glutamate carboxyl groups must be differentiated in a multistep route. The amino group remains available after neutralization for acylation or installation of an N-protecting group.

Dual Carboxyl Protection Map

OtBu and OBzl provide different chemical handles on the α- and γ-carboxyl groups. In suitable routes, tert-butyl esters are acid-labile while benzyl esters can be addressed through hydrogenolysis, creating opportunities for differential deprotection.

Orthogonality is context dependent: other protecting groups, hydrogenation-sensitive motifs, and acid-sensitive functionality in the complete molecule must be considered before choosing a deprotection sequence.

Free Amino Salt as a Synthetic Intermediate

The amino function is not Boc- or Fmoc-protected; it is present as an HCl salt. This makes the compound useful for solution-phase N-acylation, N-protection, or fragment construction after appropriate base treatment.

Procurement by Protection State

The shorthand should be read carefully: H-Glu(OBzl)-OtBu·HCl is not interchangeable with Fmoc-Glu(OtBu)-OH, Z-Glu(OMe)-OH, or diethyl glutamate. CAS 105590-97-4 is the clearest exact-entity search term.

Frequently Asked Questions

Which glutamate carboxyl group is OBzl-protected?

OBzl denotes the side-chain γ-carboxyl benzyl ester in this shorthand.

Which carboxyl group is OtBu-protected?

OtBu denotes the α-carboxyl tert-butyl ester.

Is the amino group protected with Fmoc or Boc?

No. The amino group is supplied as the hydrochloride salt and may require neutralization or N-protection in the next step.
