DESCRIPTION

[Gly22]-β-Amyloid (1-40) is a synthetic peptide variant of the amyloid β (Aβ) protein, featuring a glycine (Gly) substitution at the 22nd amino acid position (native Aβ(1-40) contains glutamate, Glu, at this site). This modification introduces structural flexibility at a critical region of the peptide, as glycine’s lack of a side chain disrupts the native hydrophobic and electrostatic interactions that drive Aβ aggregation. Aβ(1-40) is a major component of amyloid plaques in Alzheimer’s disease, and the [Gly22] variant is used in research to investigate how amino acid substitutions at position 22 affect the peptide’s propensity to form oligomers, fibrils, and toxic aggregates. By altering the conformational stability and aggregation kinetics, this variant helps scientists study the role of specific residues in Aβ misfolding, neurotoxicity, and pathological signaling. It serves as a valuable tool for understanding the molecular mechanisms of amyloidosis, evaluating the impact of genetic or post-translational modifications on Aβ biology, and developing therapeutic strategies to inhibit harmful aggregation or disrupt plaque formation in Alzheimer’s disease and related disorders.
