DESCRIPTION

[Gly22]-Amyloid β-Protein (1-42) is a synthetic peptide variant of the amyloid β (Aβ) protein, featuring a glycine (Gly) substitution at the 22nd amino acid position (native Aβ(1-42) contains glutamate, Glu, at this site). This modification alters the peptide’s structural flexibility and hydrophobic interactions, as glycine’s lack of a side chain disrupts the native conformation at a critical region involved in Aβ aggregation. Aβ(1-42) is a key component of amyloid plaques in Alzheimer’s disease, known for its high propensity to form neurotoxic fibrils. The [Gly22] variant is used in research to investigate how amino acid substitutions at position 22 affect Aβ’s aggregation kinetics, fibril morphology, and interaction with cellular targets, such as membranes or other proteins. By studying this variant, scientists aim to understand the role of specific residues in Aβ misfolding, evaluate the impact of genetic or post-translational changes on pathological processes, and develop strategies to inhibit toxic aggregation for Alzheimer’s disease therapy. Its utility lies in modeling structural perturbations and assessing therapeutic interventions targeting Aβ oligomerization or plaque formation.
