Fmoc-D-Glu(OtBu)-OH is an Fmoc-protected D-glutamic acid building block in which the side-chain gamma-carboxyl group is protected as a tert-butyl ester. The alpha-carboxyl group remains available for peptide coupling in Fmoc/tBu solid-phase synthesis.
Side-chain OtBu protection prevents unintended activation or branching through the glutamate carboxyl group and is removed under acidic global cleavage conditions. The compound is useful for D-glutamate incorporation in stereochemical peptide analogs, acidic D-residue scans and sequences designed to examine charge orientation or proteolytic recognition.

Application Scope and Assay Relevance
Primary use: Protected D-glutamate building block for peptides requiring a D-configured acidic residue with the side-chain carboxyl protected during assembly.
| Typical in vitro relevance | Typical downstream assays include receptor or enzyme SAR, charge-dependent binding, protease stability and conformational studies. D-Glu can be used to test whether side-chain orientation rather than charge alone controls activity. |
| In vivo relevance | Use in animal studies applies to the final peptide, not the protected building block. D-Glu substitution should be validated for target activity and systemic stability before in vivo interpretation. |
D-Glu(OtBu) vs D-Asp(OtBu)
The protected acidic residues differ by one methylene group. They have different CAS numbers and molecular weights, so the abbreviated names should not be treated as interchangeable.
Purchasing and QC Considerations
Confirm that the side-chain 5-tert-butyl ester is required. CAS 104091-08-9 and MW 425.47 distinguish it from Fmoc-D-Asp(OtBu)-OH and from alternate glutamate protection patterns.
Frequently Asked Questions
Which carboxyl group is protected?
The D-glutamate side-chain gamma-carboxyl group is protected as OtBu.
When is OtBu removed?
It is typically removed during acidic final cleavage.
How is it different from Fmoc-D-Asp(OtBu)-OH?
D-Glu contains one additional side-chain methylene group and has a different CAS and molecular weight.
