Pmc Versus Pbf
Fmoc-Arg(Pbf)-OH is optimized for conventional Fmoc/tBu SPPS, whereas Boc-Arg(Pmc)-OH is more relevant to Boc-based, solution-phase or historical protected-fragment routes. We consider this distinction more important than simply comparing molecular weights.
Why the Guanidino Group Needs Protection
Arginine's guanidino group is strongly basic and can complicate activation, solubility and purification if left unprotected. Pmc reduces this reactivity during protected synthesis and is removed under appropriately strong acidic conditions selected for the full molecule.
