Fmoc-β-Ala-OH is an Fmoc-protected beta-alanine building block used in peptide synthesis, beta-peptide research and flexible spacer design. Unlike alpha-alanine, beta-alanine places the amino group one carbon farther from the carboxyl group and contains no alpha stereocenter.
This simple structural difference increases backbone spacing and gives beta-alanine a distinct role in peptide-linker design, peptide conjugates and non-alpha-amino-acid sequence engineering.

Beta-Alanine vs Alpha-Alanine
Standard alanine is an alpha-amino acid in which the amino group is attached to the carbon adjacent to the carboxyl group. Beta-alanine moves the amino group to the beta carbon, adding one methylene to the peptide backbone.
This means Fmoc-beta-Ala-OH should not be treated as an L- or D-alanine substitute with the same geometry. It changes backbone length, conformational freedom and spacing between neighboring residues.
Applications in Peptide and Linker Design
Fmoc-beta-Ala-OH can be used for beta-amino-acid incorporation, peptide conjugate spacers, fluorescent or biotin linker design, lipidated peptide spacers, peptidomimetic research and construction of flexible peptide-linker architectures.
Beta-alanine can also be used as a short non-PEG spacer when a compact carbon/amide linker is preferred, although the physicochemical behavior of beta-Ala and PEG is different and they should not be assumed to be functional equivalents.
Fmoc-β-Ala-OH vs Fmoc-β-Ala-Gly-OH
Fmoc-β-Ala-Gly-OH is a preassembled beta-Ala-Gly dipeptide and introduces a longer predefined spacer. Fmoc-beta-Ala-OH provides the individual beta-alanine residue and gives the chemist more control over the next coupling partner.
