Z-Ala-Ala-OH, also written Cbz-L-Ala-L-Ala-OH, is an N-Cbz-protected L-Ala-L-Ala dipeptide with a free C-terminal carboxyl group. It provides a compact preassembled fragment for solution-phase peptide chemistry and route-development studies.
We recommend choosing a preformed Ala-Ala fragment when the route benefits from fragment condensation or when the dipeptide is repeatedly used as a defined unit. For stereochemistry-sensitive synthesis, both alanine centers should be confirmed as L/S; D-Ala-containing analogs can have the same elemental formula but different behavior.

Why Ala-Ala is a useful model fragment
Alanine has no additional reactive side-chain functionality, so an Ala-Ala fragment lets chemists evaluate backbone activation, fragment coupling, epimerization control and protecting-group compatibility without introducing extra side-chain variables. This makes the product useful as both a synthetic intermediate and a simple method-development substrate.
Cbz protection in fragment synthesis
The Z/Cbz group protects the N-terminal amine as a benzyloxycarbonyl carbamate. The C-terminal carboxyl remains available for activation. Cbz removal can be achieved by hydrogenolysis or another route-compatible method, depending on the complete molecule and the presence of hydrogenation-sensitive groups.
Procurement and QC Considerations

Frequently Asked Questions
What does Z mean in Z-Ala-Ala-OH?
Z is another name for the Cbz or benzyloxycarbonyl N-protecting group.
Is Z-Ala-Ala-OH a dipeptide?
Yes. It is an N-protected L-Ala-L-Ala dipeptide with a free C-terminal carboxyl group.
Why use a preformed Ala-Ala fragment?
It can reduce one coupling operation and provides a defined fragment for route development or larger peptide assembly.
