DESCRIPTION

[Tyr8,Nle11] Substance P is a full-length Substance P analog containing two targeted substitutions: Phe8 is replaced by Tyr and the C-terminal Met11 is replaced by norleucine.

Tyr8 Adds a Phenolic Aromatic Side Chain

Substance P normally contains Phe at both positions 7 and 8. Substitution of Phe8 with Tyr preserves the aromatic ring while introducing a para-hydroxyl group.

This provides a chemically distinct aromatic position that can be useful in ligand labeling and structure–activity experiments while leaving most of the Substance P pharmacophore unchanged.

Nle11 Removes the Methionine Oxidation Site

Met11 is replaced by norleucine, a hydrophobic non-sulfur amino acid. The substitution preserves a similar aliphatic side-chain geometry but eliminates formation of methionine sulfoxide or sulfone at the C-terminus.
