DESCRIPTION

Neurokinin A (4-10) is a seven-residue C-terminal fragment of Neurokinin A with the sequence Asp-Ser-Phe-Val-Gly-Leu-Met-NH₂. It retains the conserved tachykinin C-terminal region responsible for much of the receptor-recognition activity of the parent peptide.

The fragment provides a compact scaffold for NK2 receptor pharmacology and for determining which portions of the full Neurokinin A sequence are required for ligand activity.

The Conserved Tachykinin C-Terminus

The sequence contains the Phe-X-Gly-Leu-Met-NH₂ motif characteristic of mammalian tachykinins. Removal of the first three NKA residues produces a shorter ligand while retaining this receptor-facing region.

It therefore provides a useful starting scaffold for β-Ala substitutions, D-amino-acid antagonists and other NK2-directed analog designs.

Full-length Neurokinin A contains His-Lys-Thr before the seven-residue fragment. Comparing the two peptides can help distinguish contributions from the N-terminal extension versus the conserved C-terminal pharmacophore.
