Home Product Protected Amino Acids, Resins & Reagents Specialty Peptide Building Blocks N-Boc-Cis-Hyp-OMe

DESCRIPTION

N-Boc-Cis-Hyp-OMe is a protected cis-hydroxyproline methyl ester. It combines a Boc-protected proline nitrogen, a hydroxyl-bearing pyrrolidine ring and a methyl-ester-protected carboxyl group.

The cis-hydroxy configuration is part of the molecular identity. Hydroxyproline stereoisomers can share the same formula and nominal mass, so the product should not be selected from molecular weight alone.

Why Hydroxyproline Is Not Simply Proline Plus OH

Adding a hydroxyl group changes polarity, hydrogen-bonding capacity and ring stereochemistry. Depending on position and configuration, hydroxyproline substitutions can alter local conformational preferences in peptide and peptidomimetic structures.

The Methyl Ester Must Be Accounted For

The carboxyl group is protected as OMe, so the material is not ready for standard peptide coupling through that terminus until the ester is transformed or removed. We consider protection-state planning as important as stereochemical selection for this reagent.

Boc Protection

The ring nitrogen is Boc-protected and can be revealed under acidic conditions. Confirm the hydroxyl protection state against the product structure and lot-specific documentation before selecting route conditions.

Procurement and QC

Confirm CAS 102195-79-9, C11H19NO5, MW 245.27 g/mol and the cis-hydroxyproline stereochemical assignment. Chiral identity requires more than LC-MS.

When hydroxyproline is being incorporated as a conformational probe, evaluate the exact stereoisomer together with the complete sequence. These choices fit naturally within noncanonical peptide design and synthesis.

Frequently Asked Questions

What does Hyp mean?

Hyp denotes hydroxyproline.

Is the carboxyl group free?

No. It is protected as a methyl ester.

Can mass spectrometry distinguish hydroxyproline stereoisomers?

Not by molecular mass alone; stereochemical documentation is required.
