DESCRIPTION

Human β-CGRP, also known as Calcitonin Gene-Related Peptide II, is a 37-residue neuropeptide encoded by the CALCB gene. The mature molecule contains a Cys2-Cys7 disulfide bond and a C-terminal Phe-NH₂.

β-CGRP shares the characteristic architecture of the CGRP family and acts as an agonist at CGRP receptor systems, making it useful for receptor signaling, vascular biology and comparative α-/β-CGRP research.

β-CGRP Is a CALCB Gene Product

Human α-CGRP and β-CGRP are closely related peptides but arise from different genetic contexts. β-CGRP is encoded by CALCB, whereas α-CGRP is associated with alternative processing of CALCA.

This distinction is useful in experiments that compare CGRP isoforms because similar receptor pharmacology does not make the two peptides genetically or chemically interchangeable.

CGRP Receptor Recognition

The canonical CGRP receptor is a heteromeric complex formed by the class B GPCR calcitonin receptor-like receptor (CLR/CALCRL) and receptor activity-modifying protein 1 (RAMP1).

RAMP1 contributes directly to ligand-recognition properties of the receptor complex. The intact N-terminal disulfide ring and amidated C-terminal region of CGRP are important components of the native ligand architecture.

Human β-CGRP can therefore be used in cAMP signaling, receptor binding, ligand competition and α-/β-isoform comparison studies.

α-CGRP and β-CGRP Comparisons

The two human isoforms are highly related but not sequence-identical. Experiments designed to distinguish them should specify the exact sequence rather than using “human CGRP” as an interchangeable name.
