Fmoc-Lys(Mtt)-OH is an orthogonally protected L-lysine derivative with Fmoc on the α-amino group and 4-methyltrityl (Mtt) on the ε-amino side chain.
Mtt is deliberately more acid-labile than many global side-chain protections, allowing staged exposure of the lysine ε-amine. For a different orthogonal trigger, compare Fmoc-Lys(Dde)-OH.

Why Mtt Is Useful on Lysine
Controlled mild-acid treatment can remove Mtt while the peptide remains resin-bound, enabling site-specific side-chain modification before final cleavage.
Mtt Versus Dde
Mtt relies on acid lability; Dde is typically removed by hydrazine-based chemistry. A sequence containing other acid-sensitive groups may favor Dde, whereas hydrazine-sensitive functionality may favor Mtt. We consider this a route-level decision.
Applications After Side-Chain Exposure
The liberated ε-amine can be acylated with fluorophores, lipids, linkers, chelators or peptide branches. Selective lysine chemistry is common in probes, branched peptides and site-defined conjugates.
Procurement and QC
PubChem confirms CAS 167393-62-6, formula C41H40N2O4 and MW about 624.8 g/mol. Confirm L stereochemistry and ε-Mtt identity on the lot-specific COA.
For multi-step on-resin modification, define the exact order of Mtt removal, acylation, remaining chain elongation and final cleavage. Alan Scientific can plan these operations through custom modified peptide synthesis.

Frequently Asked Questions
What does Mtt protect?
Mtt protects the lysine ε-amino side chain.
How does Mtt differ from Dde?
Mtt is acid-labile, whereas Dde is typically removed under hydrazine-based conditions.
What can be attached after Mtt removal?
Linkers, fluorophores, lipids, chelators or other acyl groups can be introduced when compatible with the sequence.
