Fmoc-Lys(Me,Boc)-OH is a protected L-lysine derivative for introducing epsilon-N-monomethyllysine at a defined peptide position during Fmoc solid-phase peptide synthesis. The alpha amine carries Fmoc protection, while the side-chain nitrogen carries the desired methyl group plus a removable Boc group.
For histone-peptide projects, we recommend QC that confirms both sequence and modification state. LC-MS establishes mass but does not by itself exclude all methylation-state or deletion impurities. When a peptide is used for reader-domain binding or antibody specificity, small compositional errors can change the interpretation of the assay.

Why the exact lysine methylation state matters
Unmethylated, mono-, di- and trimethyllysine are distinct recognition states in epigenetic protein interactions. A peptide labeled simply “methyllysine” is ambiguous. This building block is specifically configured to yield monomethyllysine after final side-chain deprotection.
SPPS use and deprotection logic
Standard Fmoc cycles remove the alpha-amino protecting group while the side-chain Boc remains in place during assembly. The Boc group is removed during TFA-mediated final cleavage, exposing the epsilon-N-monomethyl side chain. Standard coupling methods can be used, although difficult sequences may still require optimized equivalents or repeat coupling.
