Home Product Protected Amino Acids, Resins & Reagents Specialty Peptide Building Blocks Fmoc-Gln(Xan)-OH

DESCRIPTION

Fmoc-Gln(Xan)-OH is Nα-Fmoc-Nγ-xanthyl-L-glutamine. The xanthyl (Xan) group protects the glutamine side-chain amide nitrogen during peptide synthesis while the α-amino group remains compatible with Fmoc-SPPS.

This building block is most relevant when glutamine side-chain chemistry becomes a synthesis risk rather than when a routine unprotected side-chain Gln reagent is already performing well.

Product Information

Product NameFmoc-Gln(Xan)-OH
Catalog No.AS4180
CAS No.185031-81-6
Molecular FormulaC33H28N2O6
Molecular Weight548.60 g/mol
Material / Building Block TypeFmoc-protected Nγ-xanthyl L-glutamine
Primary ApplicationsGln-containing peptide synthesis; suppression of Gln side-chain dehydration; difficult-sequence SPPS; acid-labile side-chain protection

Application Scope in Gln-Rich and Difficult Sequences

Xan-protected Gln can be useful in long, aggregation-prone or impurity-sensitive sequences where side-chain dehydration or other Gln-derived byproducts complicate purification. It provides a route option for controlling side-chain amide chemistry during activation.

Xanthyl Protection and Deprotection Window

The Xan group masks the side-chain amide nitrogen and is acid-labile. It should be evaluated against all other acid-sensitive groups in the sequence so that final cleavage and side-chain deprotection remain compatible.

Analytical Identity and Storage

Confirm CAS 185031-81-6, C33H28N2O6, MW 548.60 g/mol and Nγ-xanthyl protection. Major supplier data list sealed dry storage at 2-8°C and classify the material as H302/H315/H319.

Procurement and QC Considerations

For Gln-rich targets or routes with recurrent Gln side products, Alan Scientific can evaluate the protection strategy through custom peptide synthesis.

Frequently Asked Questions

What does Xan protect?

It protects the glutamine side-chain amide nitrogen.

Why use Xan-protected Gln?

It can reduce selected Gln side reactions during activation and chain assembly.

Is Xan removed during final cleavage?

It is acid-labile and is normally removed under an appropriately designed final deprotection regime.
