DESCRIPTION

Amyloid β-Protein (1-43) is a 43-amino acid peptide derived from the amyloid precursor protein (APP), generated through sequential cleavage by β-secretase and γ-secretase. This variant features an extended C-terminal region compared to the more commonly studied Aβ(1-40) and Aβ(1-42), with the additional three amino acids influencing its biophysical properties, such as aggregation kinetics and membrane interaction. While less prevalent in amyloid plaques, Aβ(1-43) plays a role in the early stages of APP processing and serves as an intermediate in the generation of shorter, more aggregation-prone Aβ species. In research, it is used to investigate γ-secretase cleavage specificity, the impact of C-terminal extensions on Aβ oligomerization and fibril formation, and their contributions to neurotoxicity in Alzheimer’s disease. Studying this variant helps clarify the structural basis for Aβ’s pathological behavior, supporting efforts to understand the amyloid cascade and develop therapies targeting abnormal APP processing or toxic Aβ assemblies.
