DESCRIPTION

[Pyr6]-Substance P (6-11), also written [pGlu6]-Substance P (6-11), is a six-residue C-terminal Substance P analog with the sequence Pyr-Phe-Phe-Gly-Leu-Met-NH2.

The peptide preserves the hydrophobic C-terminal region of Substance P while replacing the N-terminal Gln6 of the fragment with pyroglutamate. This compact scaffold has been used in tachykinin pharmacology and structure–activity studies and also serves as an important parent peptide for more selective analogs such as septide.

Pyroglutamate-Capped Substance P C-Terminal Fragment

The C-terminal region of Substance P contains important structural determinants for tachykinin receptor recognition. [Pyr6]-Substance P (6-11) retains the Phe-Phe-Gly-Leu-Met-NH2 region while introducing a cyclic pyroglutamate residue at the N-terminus of the fragment.

Pyroglutamate removes a free N-terminal amino group and creates a chemically distinct capped peptide. This difference should be considered when comparing [Pyr6]-SP(6-11) with the corresponding uncapped Substance P fragment.

A Parent Scaffold for Septide Pharmacology

[Pyr6]-SP(6-11) is particularly useful as a structural comparator for [Pyr6,Pro9]-Substance P (6-11), or Septide. Septide differs by the additional Gly9 → Pro substitution and displays substantially altered receptor selectivity.

We recommend including the parent [Pyr6]-SP(6-11) when studying the contribution of Pro9 because this allows the effect of the backbone-constraining substitution to be separated from the pyroglutamate modification already present in both peptides.
