DESCRIPTION

HIV-1 Tat (47–57), sequence YGRKKRRQRRR, is an 11-amino-acid, highly cationic cell-penetrating peptide derived from the basic protein transduction domain of HIV-1 Tat.

It is one of the most widely studied TAT-derived CPP sequences and is frequently used in intracellular delivery, cellular uptake, peptide conjugation, and membrane-transport research.

A Classic TAT Cell-Penetrating Peptide

Tat(47–57) contains a highly basic Arg/Lys-rich region that supports interaction with negatively charged cellular membranes.

The peptide is commonly used as a benchmark CPP when evaluating intracellular transport of peptides, proteins, fluorescent probes, nanoparticles, and other experimental cargos.

Its behavior, however, depends strongly on the experimental system. Cargo attachment can change peptide charge, hydrophobicity, uptake efficiency, and cytotoxicity.

Tat(47–57) vs Tat(49–57)

Tat(47–57):

YGRKKRRQRRR

Tat(49–57):

RKKRRQRRR

Tat(49–57) represents the shorter minimal basic motif, while Tat(47–57) retains the additional N-terminal Tyr-Gly residues.

We recommend reporting the exact Tat fragment used in an experiment rather than describing all constructs simply as “TAT peptide.”

Research Applications

Tat(47–57) is used in studies of intracellular cargo delivery, protein transduction, fluorescent labeling, nanoparticle functionalization, peptide–membrane interactions, and CPP structure–activity relationships.

Project-specific Tat derivatives can be prepared through our Custom Peptide Synthesis service.

Experimental Considerations

Cellular uptake can vary with peptide concentration, cargo size, linker chemistry, labeling position, incubation time, and cell type.

For quantitative comparisons, terminal chemistry and labeling position should remain consistent between experimental groups.

Batch identity and purity can be evaluated using analytical HPLC and MS through our Peptide Quality Control workflow.
