D-Glu5 Is Not the Same Modification as pGlu5
D-Glu5 retains an open glutamate side chain and reverses the stereochemistry of the α-carbon. It should not be confused with pGlu5, or pyroglutamate, which is a cyclic N-terminal residue found in a chemically distinct Substance P fragment commonly used as a G-protein antagonist.
This distinction matters when comparing published experiments because [D-Glu5,D-Trp7,9,10]-SP(5-11) and [pGlu5,D-Trp7,9,10]-SP(5-11) differ in sequence, molecular formula and terminal chemistry.
Multiple D-Trp Substitutions and Peptide Conformation
D-Trp substitutions invert side-chain orientation at three positions in the C-terminal pharmacophore. This changes receptor-facing topology while retaining highly aromatic indole groups.
