DESCRIPTION

Boc-Lys-OH is Nα-Boc-L-lysine. The α-amino group is Boc-protected, while the lysine ε-amino side chain remains free.

That free ε-amine is the defining feature. This material should not be treated as a routine fully protected lysine monomer for linear peptide synthesis, because the unprotected side-chain amine can participate in acylation unless the route is deliberately designed around it.

Why a Free ε-Amine Can Be Useful

Selective protection of the α-amino group leaves the side-chain amine available for derivatization. This can support preparation of ε-acylated lysine analogs, branch points, linkers, labels, or other customized lysine building blocks.

Why It Is Not a Drop-In Standard Lysine Reagent

In ordinary peptide elongation, a free ε-amine can compete with the intended backbone amine or react during activated-carboxyl chemistry. Additional protection is normally required if the side chain must remain inert throughout chain assembly.

Boc Protection and Route Design

Boc on the α-amino group is acid-labile. If the side chain is derivatized first, the newly installed ε-substituent should be chosen with the later Boc-removal and coupling sequence in mind.
