Direct Biotinylation of the Kemptide Substrate

The LRRASLG region retains the serine-containing recognition sequence used in PKA phosphorylation assays, while N-terminal biotin provides a high-affinity handle for streptavidin-based analytical formats.

This configuration can support immobilization, capture and separation of substrate or phosphorylated product without introducing an additional long-chain spacer.

Direct biotinylation changes the N-terminal chemical environment of Kemptide. We therefore recommend confirming assay performance when kinetic parameters from unmodified Kemptide are used as a reference.

Research Applications

Biotin-Kemptide can be incorporated into streptavidin plate assays, phosphorylation detection workflows, kinase activity studies and assay-development platforms where substrate capture is required.

For experiments requiring greater separation between the affinity tag and substrate sequence, Biotin-LC-Kemptide provides an alternative spacer-containing format.

Assay Design Considerations

The biotin–streptavidin interaction simplifies capture but does not itself identify phosphorylation. A compatible phosphopeptide-detection method or other assay readout is still required to distinguish phosphorylated from unmodified substrate.

For quantitative kinase studies, we recommend controlling substrate concentration, enzyme amount, reaction time and ATP conditions rather than assuming that a biotinylated derivative will reproduce the exact kinetics of unmodified Kemptide.
