DESCRIPTION

α-Neoendorphin (1-8) is an eight-residue N-terminal fragment of α-Neoendorphin with the sequence Tyr-Gly-Gly-Phe-Leu-Arg-Lys-Tyr. It retains the Leu-enkephalin-like Tyr-Gly-Gly-Phe-Leu motif together with the basic Arg6-Lys7 extension and Tyr8.

The fragment is useful for examining how removal of the Pro9-Lys10 C-terminal region changes the pharmacological and processing properties of the full prodynorphin-derived peptide.

Truncating the Prodynorphin-Derived Scaffold

Full-length α-Neoendorphin contains ten residues. Removing Pro9-Lys10 shortens the C-terminal address region while leaving the core opioid motif and Tyr8 intact.

This makes the 1-8 fragment useful for mapping how C-terminal basic and conformational features contribute to receptor preference.

Relation to κ-Opioid Pharmacology

Full-length α-Neoendorphin is a prodynorphin peptide with strong κ-opioid activity in classical receptor systems. Truncation of prodynorphin peptides can shift potency and receptor preference, so α-Neoendorphin (1-8) should be evaluated as its own molecular entity rather than assumed to reproduce the full decapeptide.

Frequently Asked Questions

How does α-Neoendorphin (1-8) differ from full-length α-Neoendorphin?

It lacks the final Pro9-Lys10 residues.

Does it retain the Leu-enkephalin motif?

Yes. Residues 1-5 are Tyr-Gly-Gly-Phe-Leu.
