DESCRIPTION

Human Hypocretin (70-98) is a 29-residue pro-orexin-derived sequence closely related to mature human Orexin B. It contains the complete Orexin B sequence followed by an additional C-terminal glycine and retains a free carboxyl terminus.

This molecular form is therefore particularly useful for studying peptide precursor processing and C-terminal amidation rather than being treated as chemically identical to mature Orexin B.

Gly-Extended Relative to Mature Orexin B

Mature human Orexin B terminates in Met-NH₂. Hypocretin (70-98) instead contains Met-Gly at the C-terminus and is not amidated.

The terminal Gly is consistent with a precursor-related form from which an amidated peptide can be generated during peptide maturation. This distinction should be retained when interpreting receptor or processing studies.

Useful for Precursor-Processing Research

The peptide can support investigations of pro-orexin organization, amidation-related processing, analytical method development and differentiation between precursor-derived and mature orexin molecular forms.

We recommend reporting the complete 29-residue sequence whenever this material is used because the names “hypocretin” and “orexin” are sometimes applied broadly to multiple processed forms.

Analytical Considerations

The sequence contains Met28, making oxidation state relevant during LC-MS analysis. The one-residue difference from mature Orexin B also creates a defined mass shift that can be used to distinguish precursor-related and amidated forms.
