Colorimetric Detection of DEVD Cleavage

Caspase-mediated hydrolysis releases free pNA from the C-terminal Asp. The liberated chromophore can be monitored near 405 nm, enabling enzyme activity to be quantified using a conventional absorbance reader.

This makes Ac-DEVD-pNA useful for endpoint assays, kinetic measurements and screening workflows where fluorescence detection is unnecessary or unavailable.

Executioner Caspase Research

DEVD-based substrates are strongly associated with caspase-3/-7 activity and are widely used in apoptosis research. They can also be processed by additional cysteine proteases under some conditions.

We therefore recommend reporting the reagent as a DEVD substrate and using biological controls when assigning cleavage to a single caspase.

Choosing pNA or AMC

pNA provides a direct colorimetric readout and simplifies instrumentation. AMC generally offers greater fluorescence sensitivity. The preferred reporter depends on assay scale, sample background and the available detection platform.
