DESCRIPTION

Z-Gly-Gly-OH, also written Cbz-Gly-Gly-OH, is an N-Cbz-protected Gly-Gly dipeptide with a free C-terminal carboxyl group. It provides a defined two-glycine fragment for solution-phase coupling and protected intermediate synthesis.

The closest Fmoc-protected counterpart is Fmoc-Gly-Gly-OH. Choosing between the two should be based on the required deprotection chemistry, not on the Gly-Gly sequence alone.

Why Cbz Protection Changes Route Planning

Cbz is commonly removed by hydrogenolysis or other reductive methods, whereas Fmoc is base-labile. Z-Gly-Gly-OH is therefore especially useful in solution-phase and protected-fragment routes where reductive N-deprotection is compatible with the rest of the molecule.

Gly-Gly as a Compact Flexible Unit

A Gly-Gly segment introduces minimal side-chain bulk and high local conformational freedom. That can be useful in short spacers or linker regions, but increased flexibility can also alter proteolytic susceptibility and the spatial relationship between attached functional groups.

Fragment Coupling Versus Stepwise Assembly

A preformed Gly-Gly fragment can simplify a convergent route, while routine SPPS may favor individual glycine additions. The decision depends on the complete sequence, protection scheme, and downstream purification plan; the SPPS practical guide provides context for stepwise resin-based assembly.

Procurement and QC

Confirm CAS 2566-19-0, formula C12H14N2O5, MW 266.25 g/mol, Cbz protection, and exact dipeptide chain length. Because nearby oligoglycine products can look similar in abbreviated catalog names, verify the structure and mass together.

For protected-fragment work, the main procurement question is whether the Cbz deprotection step is compatible with all other reducible groups in the planned intermediate.

Frequently Asked Questions

Is Z-Gly-Gly-OH the same as Fmoc-Gly-Gly-OH?

No. They contain the same Gly-Gly sequence but use different N-terminal protecting groups and deprotection conditions.

Can the C-terminal carboxyl be coupled further?

Yes. It is free and can be activated for additional amide-bond formation.

When is Cbz protection preferred?

It is useful when reductive deprotection fits the overall solution-phase or fragment-synthesis route.
