DESCRIPTION

Porcine β-Endorphin is a 31-residue endogenous opioid peptide derived from the porcine POMC/β-lipotropin pathway. It retains the conserved Tyr-Gly-Gly-Phe-Met opioid motif while containing a species-specific C-terminal sequence distinct from human, rat and several other mammalian β-Endorphins.

A Single Natural Substitution Distinguishes the Porcine Sequence

Porcine β-Endorphin differs from the camel/bovine-like sequence at position 23, where Val replaces Ile. The remaining C-terminal His27 and Gln31 residues are characteristic of several non-human mammalian forms.

This creates a useful natural structure–activity comparison in which most of the 31-residue scaffold is conserved while a single hydrophobic side chain changes.

Sequential Processing in Porcine Pituitary

Classical porcine pituitary studies showed that full-length β-Endorphin(1-31) can be generated from β-lipotropin before subsequent C-terminal processing produces shorter β-Endorphin-related products.
