DESCRIPTION

[Nle4,DPhe7] α-MSH, amide; Melanotan Ⅰ is a synthetic peptide analog of α-melanocyte-stimulating hormone (α-MSH), designed to activate melanocortin 1 receptors (MC1R) on melanocytes to induce melanin synthesis and skin pigmentation. This analog features substitutions at amino acid positions 4 (norleucine, Nle) and 7 (D-phenylalanine, DPhe) and a C-terminal amide group, enhancing its stability and receptor binding affinity compared to the native hormone. By stimulating eumelanin production, Melanotan Ⅰ promotes a tan-like pigmentation, primarily studied for its potential in photoprotection, treating pigmentation disorders (e.g., vitiligo), or as a tanning agent in research settings. While it shares melanogenic properties with Melanotan II, Melanotan Ⅰ exhibits a different receptor selectivity profile. Its use remains experimental, with safety considerations and regulatory restrictions in many regions due to off-target effects and lack of approval for clinical use, making it a valuable tool for preclinical studies of melanocortin signaling and pigmentation biology.
