DESCRIPTION

Enfuvirtide; T-20 is a synthetic 36-amino-acid peptide and the first FDA-approved HIV-1 fusion inhibitor, designed to block viral entry into host cells by targeting the gp41 glycoprotein on the HIV-1 envelope. It binds to the hydrophobic heptad repeat 2 (HR2) domain of gp41, preventing the conformational changes necessary for viral and cellular membrane fusion, thus inhibiting HIV-1 infection. Clinically, Enfuvirtide is administered subcutaneously as part of combination antiretroviral therapy (cART) for treatment-experienced patients with multidrug-resistant HIV-1, reducing viral load and improving CD4+ cell counts. In pharmacological research, it serves as a tool to investigate viral entry mechanisms, membrane fusion dynamics, and the development of novel anti-HIV agents. Its unique mechanism of action highlights its role in combating drug-resistant HIV and underscores the importance of targeting viral-host interactions in antiviral therapy.
